摘要: Eukaryotic ribosome biogenesis is facilitated and regulated by numerous factors (RBFs). High-resolution cryo-EM maps have defined the molecular interactions of RBFs during maturation, but many transient dynamic interactions, particularly early assembly, remain uncharacterized. Using quantitative proteomics crosslinking coupled to mass spectrometry (XL-MS) data from a extensive set pre-ribosomal particles, we derived comprehensive time-resolved interaction map RBF engagement 60S maturation. A novel filter that efficiently eliminates false positive integration our MS with known structural information allowed us localize 22 unmapped specific intermediates identify 9 proteins represent potentially new RBFs.
