摘要: Ecdysteroids play an important role in the growth, development, and reproduction of insects. The ecdysteroid signal is transduced through a heterodimeric complex two nuclear receptors: ecdysone receptor (EcR) ultraspiracle (USP). To investigate USP during development Grapholita molesta, (USP) cDNA was obtained from transcriptome G. molesta verified by PCR. deduced amino acid sequence (GmUSP) highly homologous to those other lepidopterans, especially Manduca sexta USP-1 isoform. Relatively high expression levels GmUSP were observed immediately prior last larval (L10) molt early pupal stage (P1). Furthermore, mRNA found midgut stage. Silencing could significantly down-regulate transcriptional level ecdysone-inducible genes result increased mortality metamorphosis prolonged prepupal duration. In RNAi experiments, down-regulated following silencing, suggesting that they under control GmUSP. addition, knockdown resulted period. Taken together, present results indicate may directly or indirectly affect molesta.
