摘要: Renal interstitial fibrosis is the most important pathological process in chronic renal failure. Previous studies have shown that poricoic acid A (PAA), main chemical constituent on surface layer of mushroom Poria cocos, has protective effects against oxidative stress and acute kidney injury. The present study aimed to investigate potential roles PAA associated molecular mechanism. NRK-49F cell line was treated with transforming growth factor-β1 (TGF-β1) or without platelet-derived factor C (PDGF-C). Cell Counting Kit-8 assay, western blotting 5-ethynyl-2'-deoxyuridine immunofluorescence staining were performed examine growth, protein expression proliferation, respectively. Data from showed 10 µM attenuated TGF-β1-induced extracellular matrix (ECM) accumulation, formation proliferation. activation Smad3 mitogen-activated kinase (MAPK) pathways also inhibited by treatment. PDGF-C reversed inhibitory fibroblast proliferation Smad3/MAPK pathway. suggested suppression ECM mediated via inhibition PDGF-C, MAPK pathways. findings not only revealed anti-fibrotic fibroblasts, but provided a new insight into prevention regulation signaling
