Nerve growth factor plays a divergent role in mediating growth of rat C6 glioma cells via binding to the p75 neurotrophin receptor.

摘要: Dysregulation of proliferation, differentiation and cell death play a major role in glial tumors, there is evidence for regulatory mechanisms involving nerve growth factor (NGF) its receptors various CNS-derived tumor lines. The aim our study was to observe the effect exogenous recombinant NGF on C6 rat glioma growth, characterize endogenous p75 neurotrophin receptor (p75) rule out whether necessary mediate NGF. Recombinant (1–100 ng/ml) applied under different serum conditions (0%, 1%, 5%) knockdown achieved by lipid-mediated antisense oligonucleotide treatment. In presence serum, had positive whereas absence produced negative number. A or increased numbers enhanced BrdU incorporation. p75-knocked down cells did not enhance serum. We conclude that (1) enhances but decreases number (2) mediated alone heterodimers containing (3) either basal levels moderate represent an autoregulatory potential cells.
