Impaired functionality and phenotypic profile of dendritic cells from patients with multiple myeloma.

摘要: Multiple myeloma (MM) is a B cell cancer characterized by clonal proliferation in the bone marrow and impaired immunity. Because MM an incurable malignancy, efficient consolidation needed urgently. Targeting clonotypic cells idiotype vaccination has proved principle to be effective indicated that future strategies, including dendritic cell-based vaccination, could suitable approach. However, as patients suffer from general immunity, which may include (DCs), careful evaluation of phenotypic traits functionality DCs necessary before vaccine can developed. This study determined number, profile myeloid plasmacytoid purified directly blood at diagnosis. A reduced number lower expression human leucocyte antigen (HLA) molecules was observed on both compared healthy controls. Also, CCR5, CCR7 DEC205 normal donors. In addition, capacity stimulate allogeneic T cytokine production decreased, suggesting these are functionally impaired. Finally, analysis samples following chemotherapy transplantation demonstrated increased HLA molecules, this time-point optimal for harvest use vaccination.
