摘要: Neurospora crassa is a filamentous fungus that grows on semisolid media by forming spreading colonies. Mutations at several loci prevent this growth. cot-1 temperature sensitive mutant of N.crassa exhibits restricted colonial At temperatures above 32 degrees C colonies are compact while lower growth indistinguishable from the wild type. Restricted due to defect in hyphal tip elongation and concomitant increase branching. We have isolated genomic cosmid clone containing type allele complementation. Sequence analyses suggested encodes member cAMP-dependent protein kinase family. Strains which we disrupted viable but display Duplication, ectopic integration promoter-containing fragment includes first one-third (209 codons) structural gene, unexpectedly resulted Our results suggest an active COT1 required for one or more events essential elongation.
