摘要: Mobilized peripheral blood precursor cells (PBPC) are used with increasing frequency to restore autologous hematopoiesis following high-dose radio-chemotherapy. The success of this method has aroused interest in the use mobilized PBPC for allogeneic transplants. This approach would eliminate need marrow aspiration and general anesthesia. In project we tested feasibility histoincompatible transplants rabbits. Adult outbred Red Burgundy rabbits were as donors, New Zealand White opposite sex recipients. One individual donor was one recipient. Conditioning consisted 10 Gy total body irradiation (TBI). Donor animals pre-treated recombinant human granulocyte colony-stimulating factor (rh G-CSF) given s.c. at μg/kg daily. Three schedules collection reinfusion 3 groups animals, each consisting 5 recipient pairs : (A) collected either on days -2, -1 0, infused once after TBI day 0 ; (B) +2, +4, +7, +9 +11 (C) consecutive days, cryopreserved 1 month followed by fresh donations +8 +11. median amount processed from 470ml (312-602) containing about 10X10 8 (5-71 x ) nucleated cells. Recipient received a 2.7 cells/kg equivalent 9.6 104 colony-forming units granulocyte-macrophages (CFU-GM)/kg (data derived Group C animals). Two 15 transplanted died toxicity, all others engrafted documented histology or chromosomal analysis. (one B C) became long-term complete chimeras. bleeding infection group) GVHD. Median survival 17 (1-23) A, 39 (12 >180 days) 18 (0 C. These results show that G-CSF-mobilized can engraft across major histocompatibility barrier. Repetitive infusions appear not intensify data support continuation recently initiated trials growth transplantation man.
