Methotrexate-induced accumulation of fluorescent annexin V in collagen-induced arthritis.

摘要: We examined the accumulation of Cy5.5-labeled annexin V in paws mice with and without collagen-induced arthritis, methotrexate (MTX) treatment, by near-infrared fluorescence imaging. Fluorescence reflectance imaging (FRI) was performed 48 hr after MTX injection at 10 min 3 Cy5.5-annexin (1 nmol dye per mouse). With arthritic paws, treatment caused a 7-fold increase intensity compared untreated 4-fold to nonarthritic MTX-treated (p < .001 each). Tissue samples were histologically for Cy5.5 TUNEL staining apoptosis. seen hyperplastic synovia mice, apoptosis showed apoptotic cells synovia. Monitoring uptake FRI provided method assessing response MTX, that readily quantitated simple instrumentation occurred before conventional measurements response.
