Rapid identification, isolation, and evaluation on anti-neuroinflammatory activity of limonoids derivatives from the root bark of Dictamnus dasycarpus.

摘要: A total of 49 limonoids derivatives were rapidly identified by UNIFI software and three new derivatives, named dasycarinone (1, DAS), isodictamdiol C (2) (3), along with nineteen known compounds, isolated from the root bark Dictamnus dasycarpus, as "Baixianpi" in Chinese. Their structures elucidated on basis spectroscopic data (UV, IR, HR-ESI-MS, NMR, CD spectra OR). All compounds tested for anti-inflammatory activities suppressing nitric oxide (NO) production lipopolysaccharide (LPS) induced BV-2 cells. DAS exhibited a strong activity IC50 value 1.8 μM. Nuclear Factor kappa B (NF-κB) luciferase assay enzyme-linked immune sorbent indicated that can suppress release inflammatory cytokines such Tumor Necrosis α (TNF-α), interleukin 6 (IL-6) via inactivating NF-κB signaling pathways. Moreover, we found obacunone-class are better than those limonin-class analyzing structure-activity relationship. Our results suggested obacunone play an important role anti-inflammation Baixianpi.
