The genes coding for the antigen 85 complexes of Mycobacterium tuberculosis and Mycobacterium bovis BCG are members of a gene family: cloning, sequence determination, and genomic organization of the gene coding for antigen 85-C of M. tuberculosis.

摘要: A gene encoding the 33-kDa secreted protein of Mycobacterium tuberculosis (antigen 85-C) was isolated and sequenced. The corresponding DNA sequence contains a 1,020-bp coding region. deduced amino acid corresponds to 340-residue consisting 46-amino-acid signal peptide 294-amino-acid mature protein. Comparison with previously described genes for 30-kDa antigen (the alpha M. bovis BCG, also called 85-B) 32-kDa antigens from BCG (antigens 85-A) indicates that three share considerable homology (70.8 77.5%) but may code distinctive epitopes. Strong differences among sequences are clearly visible upstream downstream region proteins. have been detected in genome by Southern blot hybridization type-specific probes. Furthermore, large fragments (100 1,000 kbp) separated pulsed-field gel electrophoresis showed 85 complex not clustered within bacterial genome.
