摘要: Studies with human adipose tissue have demonstrated the presence of key enzymes fat synthesis. However, long-term regulation these has not been reported. To address this issue, we used adipocytes in primary culture. Human was obtained from abdominal patients undergoing surgery. Adipocytes were isolated by collagenase digestion and cultured media supplemented 1% fetal bovine serum. evaluate metabolic activity cells, assessed following during culture : DNA pattern, cell size, glucose consumption activities for two lipogenic enzymes, fatty acid synthase (FAS) glycerol-3-phosphate dehydrogenase (GPDH). Analysis pattern showed that under above conditions did undergo apoptosis. In addition, no significant change size occurred 22 d Glucose cells also constant 60% greater 10 nmol/L insulin. Treatment insulin 3-22 increased GPDH FAS 2.8-fold, respectively, compared to without Furthermore, increase due treatment dose dependent maximal at nmol/L. Our studies show first time can be maintained viable metabolically active 2-3 wk Interestingly, remain responsive Therefore, system will allow further characterization lipogenesis useful developing pharmacological treatments obesity.
