摘要: A single amino acid substitution, from histidine to tyrosine at position 274 of the neuraminidase gene has converted Oseltamivir sensitive H5N1 influenza virus into a resistant strain. Currently, is being stockpiled in many countries potentially affected by subtype epidemic. To identify this change Oseltamivir-treated patients, method based on real-time PCR using two labeled TaqMan probes was developed for its rapid detection. In order validate method, specimen treated (Oseltamivir-resistant strain Vietnamese patient, tigers) and untreated subjects have been used study. The results thus obtained as well those derived clone selection sequencing showed that could clearly discriminate wild type H274 mutant 274Y variant. sensitivity assay low 10 copies/μl allowed detection mutation mixture mutant. Overall, with described here should be useful detecting Oseltamivir-resistant H274Y species various sources specimens high specificity. Such studies can address potential differences diagnostic outcomes between patients who develop detectable resistance retain only H5N1.
