摘要: Functional oligosaccharides, particularly konjac mannan oligosaccharides (KMOS), can regulate glucose metabolism. However, the molecular mechanisms involved in hypoglycemic effect of KMOS remain largely unknown. Here, supplementation on homeostasis was evaluated both high-fat diet (HFD)-fed C57BL/6J mice and high-glucosamine-induced HepG2 cells. remarkably ameliorated fasting blood glucose, tolerance, insulin tolerance HFD-fed mice. Abnormalities triglyceride glycogen metabolism liver induced by HFD were reversed supplementation. The signaling pathway activated KMOS, with stimulation GLUT2 membrane translocation uptake cells via AMPK pathway. Moreover, suppressed p-mTOR expression stimulated GSK-3β/CREB significantly upregulated leptin receptor downregulated PTP1B SOCS3 levels brain, a decreased serum concentration. Phosphorylation JAK2 STAT3 supplementation, while expressions Sirt1, Tfam, Pgc1-α brain elevated. Conclusively, attenuated HFD-induced dysfunction through regulation resistance resistance. This finding indicates that have potential value as an anti-hyperglycemic dietary supplement.
