Induction of cell senescence by targeting to Cullin-RING Ligases (CRLs) for effective cancer therapy.

摘要: Cullin-RING ligases (CRLs) are the biggest family of multiunit ubiquitin E3 ligases, controlling many biological processes by promoting degradation a broad spectrum proteins associated with cell cycle, signal transduction and growth. The dysfunction CRLs causes lot diseases including cancer, which meanwhile offers us promising approach to cancer therapy targeting CRLs. Recent studies have demonstrated that genetic or pharmaceutical inactivation often leads death activating multiple cell-killing pathways senescence, an emerging anticancer mechanism therapeutic agents. Here, we summarize induction cellular senescence its action, triggered specific subunits via approaches siRNA silencing, knockout as well small molecule inhibitor, exhibiting effect in vitro vivo.
