摘要: The DNA glycosylase MutY, which is a member of the Helix-hairpin-Helix (HhH) superfamily, excises adenine from mispairs with 8-oxoguanine and guanine. High-resolution crystal structures MutY catalytic core (cMutY), complex bound adenine, designed mutants reveal basis for specificity glycosyl bond cleavage chemistry. two cMutY helical domains form positively-charged groove adenine-specific pocket at their interface. Watson-Crick hydrogen partners are substituted by protein atoms, confirming nucleotide flipping mechanism, supporting specific binding orientation structurally related glycosylases.
