摘要: Reconstruction of skin requires both the dermal and epidermal equivalent skin. We have developed a reconstructed composed two compartments: (1) comprising an acellular substrate populated by foreskin fibroblasts (2) epidermis regenerated from normal human keratinocytes seeded onto equivalent. The contains type I III collagen glycosaminoglycans (GAGs) cross-linked chitosan. Fibroblasts into porous structure provide suitable to support cells. Keratinocytes attach quickly, exhibit mitotic activity form continuous stratified epidermis. After 2 weeks culture, histological sections show basal layer with cuboidal cells attached several suprabasal cell layers including stratum corneum. Transmission electron microscopy revealed membrane densification (hemidesmosomes) at dermoepidermal junction; however, lamina densa was found discontinuous this stage. noted presence lipid vesicles in spinous keratohyalin granules granular layer. differentiation complete terminal corneum containing corneocytes filled tonofilaments. Reconstructed skin, based on our chitosan-cross-linked collagen-GAG matrix is moφhologically should thus useful tool for vitro toxicological studies as well wound covering treatment patients severe burns.
