摘要: The SCL (tal-1, TCL5) gene is a member of the basic domain, helix-loop-helix (bHLH) class putative transcription factors. We found that (i) promoter for exon Ia contains potential recognition site GATA-binding factors, (ii) mRNA expressed in all erythroid tissues and cell lines examined, (iii) increases upon induced differentiation murine erythroleukemia (MEL) cells, inferred may play physiologic role differentiation. used gel shift transfection assays to demonstrate GATA motif binds GATA-1 (and GATA-2), also mediates transcriptional transactivation. To identify differentiation, we generated stable transfectants MEL K562 (a human chronic myelogenous leukemia line can differentiate along pathway) cells overexpressing wild-type, antisense or mutant cDNA. Increasing level expression two independent (F4-6 C19, 745 derivative) increased rate spontaneous (i.e. absence inducer) Conversely, was inhibited expressing either cDNA lacking domain. Our experiments suggest be target factor product serves as positive regulator
