摘要: Publisher Summary This chapter describes the chemical synthesis of fructose 2,6-bisphosphate. Fructose 2,6-bisphosphate is an allosteric activator 6-phosphofructo-1-kinase and inhibitor 1,6-bisphosphatase. The method based on 2-phosphate. treatment 1-phosphate with dicyclohexylcarbodiimide (DCC) in aqueous pyridine leads to formation cyclic 1,2-phosphate. Alkaline hydrolysis that compound yields 2-phosphate as one product other. Base-catalyzed ring opening 1,2-cyclic 6-bisphosphate (II) both 1,6-bisphosphate destroyed by heating alkali, whereas stable purified anion-exchange chromatography. 6-Phosphofructo-1- kinase activity activation 2,6- bisphosphate are determined aldolase-coupled assay. Purified rat liver used overall yield this limited primarily base-catalyzed reaction, but key purification alkali.
