摘要: By use of a mutational assay employing an octadecamer with mismatch in the center, it is shown that introduction phosphorothioate groups near 5'-end can protect against degradation by 5'-3'-exonuclease activity Escherichia coli DNA polymerase I. An optimal level protection achieved when are incorporated at least second and third internucleotidic linkages from 5'-end. However, gel electrophoretic analysis as well closer to reveals oligonucleotide not completely blocked but only slowed down.
