摘要: BACKGROUND: Adipose-derived stem cells (ADSCs) are capable of differentiating into cardiomyogenic and endothelial in vitro. We tested the hypothesis that transplantation ADSCs myocardial scar may regenerate infracted myocardium restore cardiac function. METHODS: were isolated from fatty tissue New Zealand white rabbits cultured Iscoves modified dulbeccos medium. Three weeks after ligation left anterior descending coronary artery rabbits, either a graft untreated (UASCs, n = 14), 5-azacytidine-pretreated (AASCs, 13), or phosphate buffer saline (n 13) injected infarct region. Transmural size, function, immunohistochemistry performed 5 cell transplantation. RESULTS: culture demonstrated fibroblast-like appearance expressed CD29, CD44 CD105. Five transplantation, transmural size AASC-implanted hearts was smaller than other hearts. Many differentiated cardiomyocytes. The AASCs prescar appeared more myotube-like. middle UASCs, contrast, poorly differentiated. Some participate vessel-like structures formation. All ADSC-implanted had greater capillary density region did control Statistical analyses revealed significant improvement ventricular ejection fraction, performance index, end-diastolic pressure, peak +dP/dt, two groups relative to higher -dP/dt values control, fraction +dP/dt UASC-implanted CONCLUSIONS: transplanted formed islands structures, induced angiogenesis improved 5-Azacytidine pretreatment before implantation is desirable for augmenting myogenesis. Transplantation 5-azacytidine-treated efficient preservation
