摘要: Mice were immunized with preformed complexes of dinitrophenylated haemocyanin (DNP-KLH) and anti-DNP or anti-KLH antibodies, subsequently assayed for the generation B memory (BM) cells. Complexes formed at equivalence in slight antigen excess far more effective than alone generating memory: as little 100 ng DNP-KLH-anti-DNP produced substantial cell priming. Anti-carrier anti-hapten antibodies equally effective. generated rapidly alone, adjuvant effect was not simply due to aggregation antigen. Optimal priming by required integrity Fc portion antibody: F(ab')2 antibody fragments less The capacity prime BM cells abrogated depriving mice C3. C3 also localization within splenic lymphoid follicles; made localized follicles, but efficiently those intact antibody. These results extend earlier findings (Klaus & Humphrey, 1977) strongly suggest that involves C3-dependent antigen-antibody follicles strengthen concept germinal centres are birthplace
