DNA conformation-dependent activities of human mitochondrial RNA polymerase

摘要: Mitochondrial RNA polymerase (POLRMT) is a core protein for mitochondrial DNA (mtDNA) transcription. In addition, POLRMT assumed to be involved in replication, although its exact role not yet clearly elucidated. We have found novel properties of human using reconstituted transcription system. Various lengths molecules were synthesized from templates even without defined promoter sequence, when we used supercoiled circular double-stranded as template. This promoter-independent activity was strong the promoter-dependent one. Promoter-independent conformation-dependent required TFB2M. On templates, strongly suppressed by putatively physiological amount TFAM, while inhibited lesser extent. These different inhibition patterns TFAM may important prevention random synthesis vivo. also observed on relaxed single-stranded DNA, where no longer weakly but restored addition binding protein. suggest that these could explain characteristic features mammalian mtDNA and replication.
