摘要: Cleistogenes songorica is an important perennial grass found in the pastoral steppe of Inner Mongolia. C. flourishes drought prone environments, and therefore provides ideal candidate plant system for identification drought-tolerance conferring genes. We constructed cDNA libraries from leaves roots drought-stressed seedlings. Expressed sequence tag (EST) sequencing 5664 random clones produced 3579 high quality, trimmed sequences. The average read length ESTs was 613bp. Clustering assembly identified a non-redundant set 1499 contigs, including 805 singleton unigenes 694 multi-member unigenes. resulting were functionally categorized according to Gene Ontology (GO) hierarchy using house Bioinformatic Advanced Scientific Computing (BASC) annotation pipeline. Among total 2.2Mbp EST data, 161 putative SSRs found, frequency similar that previously observed oat Arabidopsis ESTs. Sixty-three annotated as being stress responsive, which 22 genes implicated response. Using quantitative real time RT-PCR, transcripts 13 these shown be at least three fold more, or less abundant roots, with 8 increased 5 decreased relative transcript abundance. collections generated this study are valuable resource microarray-based expression profiling, functional genomics order elucidate their role, understand underlying mechanisms songorica.
