摘要: Treatment of cultured human T-lymphoid (CEM) leukemia cells with nanomolar concentrations phorbol 12-myristate 13-acetate (PMA) resulted in a reduction cell growth and the acquisition surface antigenic pattern that is common to both suppressor cytotoxic T lymphocytes. This was detected by OKT monoclonal antibodies. PMA treatment did not cause expression function but rather induced marker. marker characterized ability treated CEM suppress [3H]thymidine incorporation into phytohemagglutinin-activated peripheral blood After 4 days from either cloned or parental population 16 nM PMA, 71-98% expressed reactivity OKT3 OKT8 antibodies whereas OKT4 OKT6 less than equal 1-8% cells. The can be divided five groups based on patterns randomly isolated clones. four these were low high each fifth group resembled parent antibody after dependent time dose require replication. Acquisition also occurred 12,13-dibutyrate 13-monoacetate, 12,13-diacetate, dimethyl sulfoxide. These results indicate related agents express phenotype resembles mature lymphocyte.
