摘要: PURPOSE To investigate the development and mineralization of avian scleral ossicles using fluorescence microscopy in combination with field emission scanning electron (FESEM) energy dispersive spectroscopy (EDS). METHODS The anterior halves whole eyeballs from chickens on embryonic (E) days E10 to E21 Japanese quail E8 E17 were fixed 100% methanol for 1 min, stained Giemsa solution 5 destained distilled water 30 then viewed by epifluorescence. Propidium iodide (PI) was used detect nuclei osteocytes ossicles. FESEM EDS show areas identify differences elemental composition different regions RESULTS Using as a stain, it possible observe detailed morphology both chicken In chickens, bone microporosities first became visible at E15. Each microporosity contained single nucleus, likely that an osteocyte. amount carbon steadily decreased during embryogenesis post-hatching, while concentration oxygen showed distinct increase over this time period. Calcium phosphate levels increased gradually post-hatching stages. CONCLUSIONS A novel approach study is presented. This methodology validated studying two species, important models developmental research.
