摘要: Receptors for Advanced Glycation End Products (RAGE) and Toll Like Receptor 4 (TLR-4) have been shown to play a role in the development of Osteoarthritis (OA). We previously that knocking out RAGE mice slows disease progression articular cartilage knee. The objective this study was determine if application compound TAK-242, TLR-4 specific inhibitor, conjunction with could further attenuate disease. Destabilization medial meniscus KO Wild Type (WT mice) performed, severity OA qualitatively analyzed through two standardized scoring systems (Mankin OARSI). also performed immunohistochemistry analyze levels HtrA1, Mmp-13 Tgf-β1, known biomarkers Surprisingly, addition TLR blocker did not offer additional protection against when had knocked out, its WT exacerbated OA. conclude while blockage pathway alone is beneficial attenuation OA, hampering offers no protective benefits.
