摘要: We have recently demonstrated that induction of Th2 responses by Schistosoma mansoni egg Ag is largely due to carbohydrates on the functioning as adjuvants. Lacto-N-fucopentaose III (LNFPIII), a polylactosamine sugar, predominant carbohydrate found in S. Ag. Therefore, using neoglycoprotein, we investigated whether LNFPIII induces vivo response and functions an adjuvant. Following intranasal immunization with linked human serum albumin (HSA) (HSA-LNFPIII), BALB/c mice mounted strong produced significantly higher levels total IgE well HSA-specific IgG, IgG1, IgE. HSA-LNFPIII was over 1000-fold more potent inducing Ab production compared HSA alone. Although itself did not function epitope for either IgG or IgE, its conjugation protein essential adjuvant activity. Moreover, fucose residue crucial production. Nasal lymphocytes from immunized IL-4, IL-5, IL-10, but IFN-γ following vitro stimulation HSA-LNFPIII. In addition, these activated nasal also showed significant increase B7-2 expression B220-positive cells. Furthermore, only both i.p. s.c. induced Abs alone, suggesting activity restricted particular route immunization. These results demonstrate Lewis type can their ability induce response.
