PHD2 in tumour angiogenesis

摘要: Originally identified as the enzymes responsible for catalysing oxidation of specific, conserved proline residues within hypoxia-inducible factor-1α (HIF-1α), additional roles prolyl hydroxylase domain (PHD) proteins have remained elusive. Of four PHD enzymes, PHD2 is considered to be key oxygen sensor, knockdown results in elevated HIF protein. Several recent studies highlighted importance tumourigenesis. However, there conflicting evidence exact role tumour angiogenesis. The divergence seems because contribution stromal-derived PHD2, and particular involvement endothelial cells, vs tumour-derived PHD2. This review summarises our current understanding angiogenesis, focusing on influences vascular normalisation neovascularisation.
