Biophysical Characterization of a Riboflavin-Conjugated Dendrimer Platform for Targeted Drug Delivery

摘要: The present study describes the biophysical characterization of generation 5 poly(amidoamine) (PAMAM) dendrimers conjugated with riboflavin (RF) as a cancer targeting platform. Two new series were designed, each presenting ligand attached at different site (isoalloxazine N-3, and D-ribose N-10) varying valency. Isothermal titration calorimetry (ITC) differential scanning (DSC) used to determine binding activity for protein (RfBP) in cell free solution. ITC data shows dendrimer conjugates have KD values ≥465 nM on basis, an affinity ~93-fold lower than that riboflavin. N-3 showed greater comparison N-10 series.
