摘要: Background. Immune regulation requires antigen recognition, signaling, activation, secretion of cytokines, and effector function by lymphocytes. Although there is redundancy in the activation immune response, some cytokines simultaneously promote suppress different pathways immunity. In experiments reported here we compare cytokine gene expression within liver allografts from tolerant rats with normal isografted tissue. We also interferon-γ (IFN-γ) supernatant mixed lymphocyte cultures using peripheral blood lymphocytes stimulated against donor antigen. Methods. Orthotopic transplantations were performed cuff technique without hepatic artery revascularization. Nonisotopic situ hybridization (ISH) was used to detect localize messenger RNA specific cells Antisense DNA probes generated interleukin-2 (IL-2), IL-4, IL-10, IFN-γ. One-way set up irradiated splenocytes, collected measure IFN-γ enzyme-linked immunosorbent assay. Results. Expression IL-10 up-regulated animals versus or (p=0.0002 0.0001, respectively). localized predominantly cytoplasm hepatocytes. Levels higher proliferating naive control animals. Conclusions. hepatocytes allograft tissue after orthotopic transplantation. believe that up-regulation cross-regulates supports cytokine-mediated dysregulation, which may be a mechanism tolerance transplantation rats.
