Brinzolamide loaded core-shell nanoparticles for enhanced coronial penetration in the treatment of glaucoma.

摘要: A neurodegenerative disorder, glaucoma is a leading cause of blindness in the world. The conventional treatment strategies do not allow significant penetration drug cornea. Therefore, we prepare brinzolamide (Brz) loaded core-shell nanoparticles (NPs) to enhance coronial and thus treating glaucoma. shell NPs was composed phosphatidylserine (PS; 1,2-diacyl-sn-glycero-3-phospho-L-serine), whereas core contains Brz encapsulated brinzolamide-phosphatidylserine-polymer poly-(DL-lactic acid-co-glycolic acid)-phosphatidylserine (Brz-PS-PLGA). synthesis Brz-PS-PLGA achieved by using coaxial electrospray process (CEP), which allows preparation particles single step. size with PS brinzolamide-poly (lactic-co-glycolic) acid (Brz-PLGA) without 571 ± 27.02 nm 456 19.17 nm, respectively. charges on surface Brz-PLGA were (-) 27.45 2.98 mV 19.47 2.83 mV. transmission electron microscopy images clearly reveal as light black layer over dark PLGA core. CEP high encapsulation where percentage entrapment efficiency for 88.13 6.43%. release study conducted simulated tear fluid revealed sustained patterns from these nontoxic cells cytotoxicity studies. Further, enhanced capable significantly reducing intraocular pressure (IOP) after administration rabbit eye comparison free Brz. results suggest that coating enhances capability IOP.
