摘要: Abstract Small conductance, Ca2+-activated K+ channels (SK channels) regulate neuronal excitability. We used patch clamp to study the actions of neuroprotective drug riluzole on recombinant SK2 expressed in HEK293 cells and native SK underlying afterhyperpolarization current (IAHP) cultured hippocampal neurons. External activated whole-cell channel currents dialyzed with a Ca2+-free intracellular solution. When applied aspect membrane giant inside-out patches, enhanced by 100 nM Ca2+ reversible concentration-dependent manner; 30 μM patches sensitized activation Ca2+, resulting leftward shift curve. Riluzole also IAHP reduced spontaneous action potential frequency chemically stimulated Modulation activity may contribute its cellular, behavioral, clinical effects.
