摘要: In this study two-dimensional NMR techniques (COSY and NOESY) have been used in conjunction with one-dimensional results to complete the assignment of proton spectrum double-stranded DNA decamer, d(ATATCGATAT)2, obtain qualitative information about numerous interproton distances molecule some limited conformational dynamics. COSY NOESY measurements combined systematically assign many resonances from H1' H2',2" sugar protons specific nucleotides double helix. This method relies on fact that within a nucleotide are scalar coupled base (AH8, GH8, TH6, CH6) right-handed helices can interact simultaneously their own those adjacent (5'-3') attached its 5' side (i.e., XpA not ApX). A experiment is identify residue whereas allows neighboring be connected (linked). The CH5 CH6 immediately identified by experiment. methyl thymine residues exhibit strong through-space interbase interactions both TH6 AH8 adenine residue.
