摘要: Aim: The aim of our study was to test the feasibility High Resolution Melting (HRM) analysis for detection BRAF V600E mutation in various types fine-needle aspiration (FNA) specimens from patients with papillary thyroid carcinoma (PTC). Materials and Methods: We analyzed fresh aspirates smears eight cases PTC: three classic PTCs (CPTC), follicular variant (FVPTC), one tall cell, oncocytic PTC. DNA extraction performed using a MasterPure purification kit. isolated quantity assessed NanoDrop spectrophotometer quality tested by PCR amplification β-globin gene native electrophoresis. HRM on LightCycler 480 (Roche). amplified 15th exon gene, selected primers flank point. Results: For all cytological specimens, adequate allowed amplification. Similarly, control did not show signs degradation amplifiable gene. Four revealed mutation: two CPTCs, PTC, cell None FVPTC had this mutation. Conclusions: represents feasible reproducible molecular technique, offering new perspectives detecting FNA specimens. In study, strong association specific histological variants highly CPTC, or but negative FVPTC.
