摘要: Triplex DNA formation involving unmodified triplex-forming oligonucleotides (TFOs) is very unstable under physiological conditions. Here, we report a novel strategy to stabilize both purine and pyrimidine motif triplex within the rat alpha1 (I) collagen gene promoter physiologically relevant conditions by poly(L-lysine)- graft -dextran copolymer. Using an in vitro electrophoretic mobility shift assay, show that copolymer almost completely abrogates inhibitory effects of concentrations monovalent cations, particularly potassium ion (K+), on low guanine-rich TFO. Of importance, pH dependency cytosine-rich TFO also significantly overcome Finally, triplex-stabilizing efficiency remarkably higher than other oligocations, like spermine spermidine. We suggest ability salt will be cue for further progress antigene strategy.
