摘要: We have previously cloned a large chondroitin sulfate proteoglycan (versican) from human fibroblasts. The primary sequence shows that the N terminus contains homology with known hyaluronate-binding molecule, suggesting versican can bind hyaluronate. To test this hypothesis we reconstructed full-length cDNA and fragment encoding transfected Chinese hamster ovary cells mouse 3T3 fibroblasts, respectively, these constructs. make shown to be by enzymatic immunologic analysis. No corresponding was seen in control cells. Using hyaluronate affinity chromatography, show recombinant specifically binds does not heparin or sulfate. fibroblasts 78-kDa truncated form of also related polysaccharides, showing binding activity resides at versican. is substrate-concentration dependent time competed unlabeled dissociation constant for determined 4 x 10(-9) M.
