摘要: This study reports on a cytotoxic toxin derived from the venom of crown-of-thorns starfish Acanthaster planci (CAV). The protein was isolated through both ion-exchange and gel-filtration chromatography, characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) mass spectrum analyzes. CAV identified as plancitoxin I protein. mechanistic role explored in human malignant melanoma A375.S2 cell death. results indicated that after incubation with toxin, cells significantly decreased viability increased lactate dehydrogenase (LDH) level dose-dependent manner. assays promoted reactive oxygen species (ROS) production, induced nitric oxide (NO) formation, lost mitochondrial membrane potential (ΔΨm) inter-nucleosomal DNA fragmentation cells. molecular cytotoxicity tested evaluation apoptosis/necrosis ratio double staining annexin V-FITC propidium iodide (PI) assay. suggested effect via apoptotic procedure, may be associated regulation p38 pathways.
