摘要: Abstract Totipotent tissue cultures of maize (Zea mays L.) previously have been initiated from various explant tissues, including immature tassels. To obtain high frequencies culture initiation tassels, we recommend the use tassels between 1 and 3 cm long, cut into 1-mm sections, incubation under low light on Murashige Skoog (MS) medium containing 1.0 mg/1 2,4-dichlorophenoxyacetic acid (2,4-D). Callus with opaque white structures resembling embryonic scutella was classified as totipotent (capable plant regeneration). Frequencies which produced one or more these ranged 0 to 100% among 13 genotypes. Ten genotypes at least 20% their explanted Not all were capable producing plants would grow maturity. The responses reciprocal F1 parental lines suggested that partially dominant nuclear genes control initiation. No evidence cytoplasmic maternal effects found. Within genotypes, embryos in vitro similar qualitatively quantitatively, thus appeared be genetic control. explants permits selection specific via markers particular phenotypes prior
