Polygonatum cyrtonema lectin induces apoptosis and autophagy in human melanoma A375 cells through a mitochondria-mediated ROS–p38–p53 pathway

摘要: Polygonatum cyrtonema lectin (PCL), a mannose-binding lectin, has been reported to induce cytotoxicity and apoptosis. Herein, we demonstrated that PCL-induced apoptosis autophagy in A375 cells. The apoptotic mechanism was PCL treatment regulated Bax, Bcl-xL Bcl-2 proteins, leading mitochondrial depolarization, cytochrome c release caspase activation. Subsequently, found abrogated glutathione antioxidant system induced mitochondria generate ROS accumulation, resulting p38-p53 Moreover, confirmed the ROS-p38-p53 pathway involved autophagy. In conclusion, these results indicate induces via mitochondrial-mediated pathway.
