摘要: Understanding the mechanisms regulating development of cereal seeds is essential for plant breeding and increasing yield. However, analysis challenging owing to minute size, liquid character some tissues, tight inter-tissue connections. Here, we demonstrate a detailed protocol dissection embryo, endosperm, seed maternal tissues at early, middle, late stages barley development. The based on manual tissue using fine-pointed tools binocular microscope, followed by ploidy analysis-based purity control. Seed embryos are diploid, while endosperm triploid tissue. This allows monitoring sample flow cytometry. Additional measurements revealed high quality RNA isolated from such samples their usability high-sensitivity analysis. In conclusion, this describes how practically dissect pure developing grains cultivated potentially also other cereals.
