Abatement of neurobehavioral and neurochemical dysfunctions in cerebral ischemia/reperfusion injury by Tetrapleura tetraptera fruit extract.

摘要: Abstract Ethnopharmacological relevance Tetrapleura tetraptera Taub. (family Fabaceae), is generally found in the lowland forest of tropical Africa. Its leaves and fruits are traditionally used West Africa for management brain disorders. Aim study This evaluated effect methanol fruit extract (TT) on bilateral common carotid artery occlusion-induced cerebral ischemia/reperfusion (I/R) injury male Wistar rats. Materials methods Rats pretreated with TT 7 days before a 30 min occlusion reperfusion 24 h were assessed neurobehavioural deficits. Cortical, striatal hippocampal oxidative stress, pro-inflammatory events, electrolyte imbalance neurochemical dysfunctions, as well histopathological alterations, also evaluated. HPLC-DAD analysis was performed to identify likely compounds contributing bioactivity extract. Results reduced I/R-induced behavioral deficits ameliorated stress by restoring glutathione level, increasing catalase superoxide dismutase activities, reducing both lipid peroxidation xanthine oxidase activity brain. attenuated I/R-increased myeloperoxidase lactate dehydrogenase activities disturbances Na+ K+ levels. Alterations elicited I/R Na+/K+ ATPase, complex I, glutamine synthetase, acetylcholinesterase, dopamine metabolism abated pretreatment. prevented histological changes hippocampus. revealed presence aridanin, marker compound tetraptera, other phytochemicals. Conclusions These findings indicate that has protective potential against stroke through modulation redox imbalances, attenuation neurotransmitter dysregulation dysfunctions. could be promising source discovery bioactives therapy.
