Production and Characterization of Hybridoma Antibody to Native Human Type II Collagen

摘要: Summary Using a sensitive RIA for collagenous proteins, we have applied series of consecutive selection steps to construct hybridoma that produces antibody against native human Type II collagen. The consisted of: 1) hybrid cell line selection; 2) co-selection IgG class immunoglobulin reactive the immunizing antigen; 3) anticollagen by complete inhibition binding collagen-coated plates prior specific collagenase digestion; 4) anti-helical heat-denatured plates; 5) clonal 6) absence crossreactivity 5–7 additional collagen types; and finally 7) yielding indirect immunofluorescent staining cartilage matrix, but not adjacent bone or perichondrium. Crossreactivity assays other types revealed little crossreactivity. In addition, demonstrated inhibits can be recovered from antigen-antibody complexes; radio immunoassay developed; is useful studies matrix.
