摘要: We improved the analytical method for detection of ascorbic acid in plasma by high-performance liquid chromatography (HPLC) with an electrochemical detector (ECD) to be more selective and rapid than protocol previously used. Main improvements are as follows. Applied potential ECD obtain maximal response was +450 mV versus Ag/AgCl, but it reduced +350 mV. In that condition, uric did not respond ECD, only detected. EDTA contained sample extraction/stabilizing solution gave peak after acid. The addition (0.2 mM) mobile phase eliminated peak. These two chromatogram which appeared from acid, shorten run time. Ascorbic unstable even though treated methanol/EDTA: decreased 1.5 h at 4 degrees C. However, sample, placed -14 C until analysis performed, reliable value least up 6 h. data obtained HPLC-ECD consistent those hydrazine method.
