摘要: Somatic embryogenesis can be synchronized by enriching carrot (Daucus carota L.) suspension cultures for small, dense clusters of cells termed proembryogenic masses (PEMs). Gene-expression programs PEMs were compared with those embryonic and mature tissues in-vitro translation representative mRNA populations nucleic-acid hybridization. Analysis invitro-translated polypeptides two-dimensional polyacrylamide gel electrophoresis revealed striking similarities between the PEM torpedo-stage embryos; substantial differences, however, observed when products torpedo embryos hypocotyls leaves. Northern blots RNA isolated from PEMs, staged embryos, hybridized cDNA probes Dc3, Dc5 Dc13; these recombinants represent mRNAs that are regulated during somatic embryogenesis. The pattern expression embryo-regulated transcripts was similar in but differed other tissues. These results indicate many molecular processes already established presence auxin. Additional experiments utility Dc3 as a marker acquisition embryogenic potential.
