Somatic pairing of chromosome 1 centromeres in interphase nuclei of human cerebellum.

摘要: Interphase nuclei isolated from paraffin-embedded tissue of four normal brains were hybridized with biotinated repetitive DNA probes specific for the (peri) centromeric regions chromosomes 1 and 7. Hybridization results visualized a peroxidase-DAB system after which number signals per nucleus was counted using bright field microscopy. Using probe chromosome 7 (p7t1), both cerebral cerebellar samples showed 2 spots in 82% 83%, respectively, nuclei. In situ hybridization (pUC1. 77) two 69% samples, pUC1.77 resulted only one large spot cells. The average size signal about 1.6 times as that signals. These observations suggest single cells is not result monosomy but it reflects somatic pairing centromeres. Based on fraction spot, small granular neuron most likely candidate. difference between indicates this cell-type-dependent phenomenon.
