摘要: Abstract The photochemical reaction of MgADP-vanadate with the active site myosin has been used to place a serine at binding for gamma-phosphate ATP. Irradiation subfragment 1 transition state-like complex UV light specifically photooxidizes hydroxyl group residue an aldehyde (Cremo, C. R., Grammer, J. C., and Yount, R. G. (1988) Biochemistry 27, 8415-8420). Reduction photooxidized Na-B3H4 gave only 3H-labeled serine. Here, subsequent extensive proteolytic digestion trypsin thermolysin yielded two peptides, both which contained sequence Gly-Glu-Ser-Gly-Ala-Gly-Lys-Thr, in all 3H was associated This is conserved heavy chains sequenced date corresponds residues 178-185 rabbit chain (Tong, S. W., Elzinga, M. (1983) Biol. Chem. 21, 13100-13110). These results Ser-180 gamma-phosphate-binding ATP indicate that glycine-rich loop around provides essential elements phosphate-binding molecules. Such role previously suggested based on common Gly-X-X-X-X-Gly-Lys-Thr/Ser, found many other nucleotide-binding enzymes (Walker, E., Saraste, M., Runswick, H., Gay, N. (1982) EMBO 1, 945-951).
