摘要: Establishment of cell polarity is essential for processes such as growth and division. In fission yeast, well other species, factors travel at the ends microtubules to cortical sites where they associate with membrane subsequently maintain a polarized activity pattern despite their ability diffuse in membrane. this chapter we present methods establish an vitro system that captures features process. This bottom-up approach allows us identify minimal molecular requirements microtubule-based polarity. We employ microfabrication techniques combined surface functionalization create rigid chambers affinity proteins, microfluidic shape emulsion droplets functionalized lipid boundaries. Preliminary results are shown demonstrating properly organized microtubule cytoskeleton can be confined these spaces, proteins traveling growing delivered
