摘要: A cell progressing through the cycle encounters various checkpoints in G1 that can lead to an exit from cycle. This ensures cells only proliferate when destined do so. Cancer, however, is characterized by uncontrolled proliferation and cancer thus bypass these checkpoints. In this chapter, will be scrutinized with emphasis on how alterations their evasion. The molecular core driving central all consists of CDKs, cyclins, activating enzymes, CKIs. Shortly after mitosis, meet a first checkpoint may direct them apoptosis presence cellular stress. After passing checkpoint, restriction point (R), which monitors growth factors pRb plays role. Two halt cycle beyond R induce or senescence: classic p53-dependent stress recently iden-tified energy checkpoint. contrast absence factor signa-ling, hypermitogenic signaling also result arrest senescence. seems mechanism prevent tumorigenesis. Finally, role reactive oxygen species control phase considered, possible involvement arrest.
