摘要: Information is presented concerning the overall arrangement of plastid DNA (ptDNA) in plastids approximately 100 spp. eukaryote algae, representing all classes. The three-dimensional ptDNA was assessed by study both living and fixed material, stained with fluorochrome 4′,6-diamidino-2-phenylindole (DAPI), using phase fluorescence microscopy. widespread occurrence two major types configuration known from prior electron microscopy studies confirmed. These are (1) densities (nucleoids) variable size morphology, scattered throughout plastid, (2) a ring nucleoid, beaded or unbeaded, lying just within girdle lamella. Type 1 characteristic Rhodophyta, Dinophyta, Chlorophyta, Cryptophyta, Prymnesiophyceae Eustigmatophyceae (with one exception). 2 Phaeophyceae, Bacillariophyceae, Raphidophyceae, Chrysophyceae (except silicoflagellates organisms such as Synura Dinobryon), Xanthophyceae exception Vaucheria three genera to lack lamellae, Bumilleria, Bumilleriopsis, Pseudobumilleriopsis). Some these exceptional forms, well Euglenophyta, have configurations not previously recognized. In observed, single could be interpreted being continuity. This character appears stable under varied conditions growth at differing stages life cycle, where examined, has confirmed reclassification made on other grounds several taxonomic entities. It also revealed new questionable classifications. Since DAPI staining far simpler than serial sectioning for revealing architecture, use technique may valuable future numerous organisms, help their identification an aid unravelling affinities. light endosymbiont hypothesis, characters require great attention those remainder cell.
