Cloning and Initial Analysis of the Human Multidrug Resistance-Related MVP/LRP Gene Promoter

摘要: The lung resistance-related protein (LRP) was identified as the human major vault (MVP), and is overexpressed in various multidrug-resistant cancer cell lines clinical samples. We characterized DNA sequences upstream to transcription initiation site of MVP gene non-small line SW-1573. A 1.9-kb a shortened 0.7-kb fragment 5'-upstream genomic region show strong promoter activity chloramphenicol acetyltransferase (CAT) reporter assays. TATA-less contains an inverted CCAAT-box Sp1 located near p53 binding motif. An alternative 3'-splice intron 1 results splicing variant within 5'-untranslated mRNA.
